The goal of pharmacogenomics research is to get genetic polymorphisms that underlie variation in drug response. Increasingly,
pharmacogenomics research involves large numbers of patients and therefore the application of latest technologies and methodologies to enable discovery. The Pharmacogenomics Research Network (PGRN) has become a community-driven network of investigators spanning scientific and clinicaldisciplines. Here we are highlighting the activities and kinds of resources that enable PGRN members to reinforce and translational research and drive basic in pharmacogenomics.
Pharmacogenomics is that the study of how genetic variation influences responses to drugs. This includes how genetic variants affect drug metabolism, efficacy and toxicity, with the goal of improving and personalizing drug therapy. The most prevalent drug-metabolizing enzymes (DME) are the Cytochrome P450 (CYP) enzymes. The term Cytochrome P450 was coined by Omura and Sato in 1962 to describe the membrane-bound, heme-containing protein characterized by 450 nm spectral peak when complexed with carbon monoxide.The human CYP family consists of 57 genes, with 18 families and 44 subfamilies. CYP proteins are conveniently arranged into these families and subfamilies on the basis of similarities identified between the amino acid sequences.
