Chromatography ( HPLC) is used mainly for component isolation , identification, and qualitative & quantitative analysis in a sample mixture. For semi-volatile or non-volatile organic compounds this is commonly used. It is a form of column
chromatography but is highly sensitive compared to column
chromatography and extremely automated. Unlike the solvent used in column
chromatography under gravity through a tube, the HPLC technique uses high pressure up to 400 atm.Due to the pressure and the wider surface area of the packing material in the column which is accommodated due to the smaller particle size, it makes a quicker and better separation than the column chromatography.
