Experimental-Genomics Review Articles
The research focuses on developing molecular methods that allow large-scale experimental genomics of DNA, RNA , and proteins in both bulk and single cell samples. Massive DNA barcoding of emulsion droplets is a method which has facilitated high-throughput investigation of a cell's functional constituents. In this method, barcodes of DNA are linked to
molecules of DNA (or cDNA) in droplets and sequenced using a platform of massively parallel
sequencing (MPS). The sequenced DNA
molecules are each traced back to a share droplet that helps individual
cells to examine
mutations and profile transcripts. The group is also working on a new method for the DNA-assisted proteomics based on droplets. The project's main focus is on a novel principle for simultaneous detection and quantification of proteins in complex samples, where a combination of immunorecognition (DNA-labeled antibodies) and massively parallel
sequencing of DNA are applied. Previously, the research group developed trinucleotide threading (TnT), protease-mediated allle-specific extension (PrASE) and apyrase-mediated allle-specific extension (AMASE) to evaluate variations in DNA and to profile expressed genes in a multiplex mode.
