Tissue Transplantation Open Access Articles
Tissue or cell
transplantation has been an extremely valuable technique for studying developmental potential of certain cell population, dissecting cell-environment interaction relationship, identifying stem cells, and many other applications. One key specialized prerequisite for performing
transplantation examine is that the capacity of recognizing the transplanted giver
cells from the endogenous host
cells and following the benefactor
cells after some time. Zebrafish has emerged as a superb
model organism for performing
transplantation assay, because of the transparency of embryos during development and even certain adults. Using transgenic techniques and fast-evolving imaging technology, fluorescence-labeled donor
cells can be easily identified and studied in vivo. In this chapter, we'll first discuss the rationale of various sorts of zebrafish
transplantation in both embryos and adults, then specialize in detailed methods of three types of transplantation: blastula/gastrula
transplantation for mosaic analysis, somatic cell transplantation, and
tumor transplantation. Tissue
transplantation into the consideration was first embraced a little more than 100 years ago.720 it's as of late, however, that cell therapeutics turned out to be adequately comprehended to legitimize endeavors to treat RP. Such photoreceptor rescue in the RCS rat has also been documented using human fetal RPE cells,722 nonfetal human RPE,723 and most recently using human induced pluripotent stem cell-derived RPE.724 Long-term effectiveness requires adjunctive immunosuppression .722 Although most efforts in RPE
transplantation have been directed toward macular diseases such as age-related macular degeneration,725–727 this methodology may be useful in RP due to RPE-specific mutations.728,729 Current tissue
transplantation add RP though is concentrated on replacing lost photoreceptors.
Cells from three different sources lead this work: adult-derived human stem cells; human embryonic stem cells; and human-induced pluripotent stem cells.
